    1,3-Diisopropylcarbodiimide (DIC) is an organic compound with the formula (i-PrN)₂C. It is a colorless to pale yellow liquid with a pungent odor, widely recognized as a versatile and efficient coupling reagent in organic synthesis, particularly in peptide chemistry. Its primary application is as a dehydrating agent for the formation of amide bonds, especially in solid-phase peptide synthesis (SPPS). DIC, often used in conjunction with additives like 1-hydroxybenzotriazole (HOBt) or Oxyma Pure, activates carboxylic acids to form O-acylisourea intermediates, facilitating coupling with amines. Compared to other carbodiimides like DCC, DIC is favored because its byproduct, N,N'-diisopropylurea, is soluble in many organic solvents and can be easily removed, simplifying purification. Beyond peptide synthesis, DIC is also employed in the synthesis of esters, anhydrides, and other compounds requiring dehydration conditions. It is a key reagent in the preparation of various pharmaceuticals and bioactive molecules. Handling requires caution as it is a lachrymator, a skin and respiratory irritant, and moisture-sensitive.
