    Fludarabine phosphate is a fluorinated purine nucleoside analog and the phosphate salt of fludarabine. It is a prodrug that is rapidly dephosphorylated in plasma to its active metabolite, fludarabine arabinoside 2-fluoro-ara-A (F-ara-A), which is subsequently phosphorylated intracellularly to the active triphosphate form (F-ara-ATP). This compound is a potent antineoplastic agent primarily used in the treatment of hematologic malignancies. Its main mechanism of action involves inhibition of DNA synthesis. F-ara-ATP competitively inhibits DNA polymerases, ribonucleotide reductase, and DNA primase, leading to termination of DNA chain elongation and induction of apoptosis in rapidly dividing cells. It is a key chemotherapeutic drug for chronic lymphocytic leukemia (CLL) and is also used in conditioning regimens prior to hematopoietic stem cell transplantation. Fludarabine phosphate is administered intravenously due to its poor oral bioavailability. It is associated with significant immunosuppression, increasing the risk of opportunistic infections, and requires careful patient monitoring. Its development and use represent a significant advancement in the treatment of lymphoid cancers.
